I went on our Annapurna Sanctuary trip about a year an a half ago and loved every minute -even the tough bits (and there were some). The trip grading is "Moderate", but this doesn't mean there will be no hard bits. The parts people found most draining were the staircases - huge stone staircases that go up an entire steep hillside. Everyone had a red face at the top, but they all made it.
There is a change to this trip for 2001. It will now be teahouse (lodge) based rather than a fully serviced trek. Also the dates have been scheduled so that the Sanctuary can be tacked on to the end of the Annapurna Circuit trek.
Gates of Mustang is along the most visited section of the Annapurna Circuit, which means it has the best trailside accommodation, but you will see plenty of other trekkers.
Awesome views on both trips unless you are unlucky with the weather.
If you are new to trekking, buy some good boots and break them in. Get fitter - no matter how fit you are - as you will enjoy it more.
Other than that, just enjoy the trip. In trekking getting there is the fun!
